[体外研究]

Direct Red 80(天狼星红)在组织病理学中有两个重要用途：从饱和苦味酸水溶液中染色胶原蛋白和从碱性溶液中染色淀粉样蛋白。染色结构为红色[1]。激光共聚焦显微镜(CLM)也用于评估直接红80染色后胶原的产生和分布。Direct Red 80对不同类型的胶原蛋白没有特异性。样品在dH2O中洗涤,用0.2%磷钼酸(PMA)溶液预处理40分钟,然后在0.1%直接红80在饱和苦味酸水溶液中稀释的溶液中培养30分钟。最后,每个样品在0.01 M HCl中洗涤2分钟。PMA用于制造无色细胞质,HCl用于去除非特异性染色[1]。

以对氨基苯磺酸、邻氨基苯磺酸、J 酸和光气为原料，首先将对氨基苯磺酸重氮化，再与邻氨基苯磺酸偶合，然后进行第二次重氮化，与 J 酸偶合，经与光气缩合得产物。经盐析、过滤、烘干、粉碎得成品。。
