[体内研究]

黄尿酸是推定的内源性II组代谢型谷氨酸受体激动剂,用于丘脑的感觉传递。当应用离子电渗疗法和静脉注射时,可以发现黄嘌呤酸(XA)对感觉的抑制,与其他II组mGlu受体激动剂相似,在生理感觉刺激下减少VB中丘脑网状核引起的抑制。此外,假定黄尿酸可能是mGlu受体的第一个潜在的内源性变构激动剂。由于II组受体和犬尿氨酸代谢途径都与精神分裂症的病理生理学密切相关,因此黄嘌呤酸在抗精神病药物研究中起着关键作用,因为这种潜在的内分泌代表了这两种生物学参数的一致性和一类新的II类。 mGlu受体配体[1]。

[动物实验]

大鼠[1]在整个研究中使用雄性Wistar大鼠（265-495g，n = 27）。在整个实验中，监测心电图和脑电图（EEG）。使用外桶进行离子电渗药物应用，其中一个外桶填充1M NaCl，用于每次均衡电流。剩余的外桶根据需要含有下列物质之一：NMDA（50 mM，pH8.0在150 mM NaCl中），LY354740（5 mM，pH8.0在75 mM NaCl中），黄尿酸（5 mM，pH8.0）在75mM NaCl中，LY341495（5mM，pH8.5在75mM NaCl中），作为Nat盐，以阴离子形式喷射，和LY487379（1mM，pH6.0，在1％二甲基亚砜，75mM NaCl中），作为阳离子喷射。通过使用与喷射电流相反极性的保持电流（15-25nA），防止所有化合物扩散出移液管[1]。
