Biological Activity

[Description]:

Clofibride is an orally active hypolipaemic agent of pchlorophenoxyisobutyric type. Clofibride has weak toxicity and marked hypocholesterolaemic and hypotriglyceridaemic activity. Clofibride is converted into 4-chlorophenoxyisobutyric acid (CPIB) and 4-hydroxy-N-dimethylbutyramide (HMB) in vivo[1].

[In Vivo]

Clofibride (oral, 400; 500 and 1000 mg/kg; single) can rapidly and massively resorb and hydrolys into 4chlorophenoxyisobutyric acid (CPIB) and 4-hydroxy-N-dimethylbutyramide (HMB) [1]. Animal Model: Male CD rats, male and female beagles, olive baboon monkeys[1] Dosage: 400, 1000 mg/kg (rats); 500 mg/kg (dogs and monkeys) Administration: oral, single Result: HMB is rapidly metabolized and the half-life of CPIB, the main active metabolite, in the serum is about 12 h in the rat, 43 +9 h in the dog and 6t 1 h in the baboon.
