缓激肽系统是一种基于组织的系统,具有强大的心血管和肾脏作用。为了研究该系统的调节,一种高灵敏度的氨基末端定向放射免疫分析,使用高效液相色谱法,可以测量缓激肽-(1-7),缓激肽-(1-8)和缓激肽-(1- 9),是开发的。与羧基末端定向的放射免疫测定一起,表征大鼠肾脏和血液中的缓激肽。肾脏中主要的缓激肽是缓激肽-(1-9)(~100 fmol/g湿重组织)和缓激肽-(1-7)(~70 fmol/g),低水平的缓激肽-(1- 8)(~8 fmol/g)和缓激肽-(4-9)(~12 fmol/g)可检测;缓激肽-(2-9)和缓激肽-(3-9)低于检测限。在血液中,缓激肽-(1-9)的水平非常低(~2 fmol/ml),而其他缓激肽则低于检测限。血管紧张素转换酶(ACE)抑制剂培哚普利的施用与缓激肽-(1-8)和缓激肽-(1-9)的肾水平增加约两倍和缓激肽-(1-7)/的减少相关。缓激肽-(1-9)比率。氨基末端定向放射免疫测定也适用于心脏,主动脉,棕色脂肪组织,肾上腺,肺和脑。对于这些组织,缓激肽-(1-7)和缓激肽-(1-9)具有相似的丰度(16-340fmol/g),具有较低水平的缓激肽-(1-8)[1]。
