[ 靶点 ]

[体外研究]

在浓度为0.01至300μM时,cirsiliol引起大鼠离体回肠的浓度依赖性松弛。 Cirsiliol可抑制Ca2 +内流,但可刺激细胞内储存的Ca2 +释放[1]。用鼠李素或cirsiliol处理可通过抑制辐射诱导的Notch-1表达来减少NSCLC细胞的增殖[2]。

[体内研究]

在异种移植小鼠模型中,与单独照射相比,通过辐射和鼠李糖素或西司它林的组合处理显着降低了肿瘤体积[2]。

[细胞实验]

将NSCLC，NCI-H1299，NCI-H460，WI-26 VA4和MRC-5细胞系暴露于单剂量的γ射线。然后用溶于DMSO的鼠李糖和cirsiliol（5,10,15,20,25μM）处理细胞4小时[2]。

[动物实验]

给小鼠[1] BALB / c无胸腺裸鼠注射2×106个NCI-H1299细胞。当肿瘤获得200mm 3的最小体积时，每天腹膜内施用DMSO或Cirsiliol（200μg/ kg体重），持续25天。每周一次用10Gy照射动物，持续3周。在第25天，切除肿瘤并进行进一步分析[1]。
