[体内研究]

在给予5'-N-乙基甲酰胺基腺苷(NECA)后,每次获得的可卡因输注的平均数量以剂量依赖性方式显着降低[5'-N-乙基甲酰胺基腺苷(NECA)：F(4,12)= 14.9; P <0.001]。给予5'-N-乙基甲酰胺基腺苷(NECA)[F(4,12)= 16.1; P <0.001]导致潜伏期显着增加,高于载体治疗获得的值[1]。每天ip注射0.3mg/kg /天的5'-N-乙基甲酰胺基腺苷(NECA)两周,降低糖尿病大鼠的丙二醛(MDA)水平,但不影响对照大鼠。每日用NECA治疗(0.3mg/kg /天,腹腔注射2周)可降低糖尿病大鼠中糖尿病诱导的肿瘤坏死因子(TNF)-α和白细胞介素(IL)-18的基因表达,但不影响对照大鼠。每日ip注射0.3mg/kg /天的5'-N-乙基甲酰胺基腺苷(NECA)两周也会阻断糖尿病大鼠JNK MAPK的活化,但不影响对照大鼠[2]。

[动物实验]

雄性Wistar大鼠用于该实验。这些动物的重量在350至425克之间。它们在12小时光照/黑暗循环下单独安装在悬挂式线笼中。在4只大鼠中测试了5'-N-乙基甲酰胺基腺苷（NECA）的作用。在该实验中，在测试开始前15分钟腹膜内施用5'-N-乙基甲酰胺基腺苷（NECA）（5,7.5,10,20μg/ kg）或载体（盐水）。按随机顺序交叉设计给予5'-N-乙基甲酰胺基腺苷（NECA）。在大多数情况下，动物用相同的剂量进行两次测试[1]。
