用作医药、农药、染料中间体
【用途二】
本品可制成2,4-二氯氟苯,后者是合成环丙沙星、培氟沙星、二氟沙星等喹喏酮类抗菌药的起始原料。本品也是一种重要的分析试剂,用来鉴定有机化合物中的氨基,尤其是用于检定肽及蛋白质末端氨基酸的氨基。检定时,2,4-二硝基氟苯与肽链的游离氨基作用,生成2,4-二硝基衍生物。将其水解后,末端氨基酸的N-(2,4-二硝基苯基)衍生物常为亮黄色结晶,易与其他氨基酸分离。用该方法结合其他方法,可确定蛋白质或多肽氨基端碳链的结构,称桑格法。本品也用于测定酚和咖啡,以及醛糖分析。

2,4-二硝基氟苯制备
1.以2,4-二硝基氯苯与无水氟化钾为原料,经亲核取代反应而得。
2.将无水氟化钾,二甲基亚砜和适量阻聚剂混合,控料加热至120℃,然后加入2,4-二硝基氯苯 [ 各反应物的配比为2,4-二硝基氯苯∶无水氟化钾∶二甲基亚砜∶阻聚剂=1.0∶1.8∶ 3.4∶0.1( 摩尔比)]。维持反应温度在110~120℃( 严格控制反应温度,以防爆炸) ,反应3h:
反应结束后迅速冷却至室温。抽滤除去氟化钾残渣,滤液中加水洗涤,静置后分去水层(可回收二甲基亚砜),油层由水洗涤2次,于2.73kPa下减压蒸馏,收集176~180℃馏分,即为成品2,4-二硝基氟苯。
