In Vitro 9-β-D-Arabinofuranosylguanine 通过其 5 ' -三磷酸腺苷 Ara-GTP 在细胞内的积累引起细胞毒性,Ara-GTP 抑制 DNA 聚合酶和核糖核苷酸还原酶,并入 DNA,终止 DNA 链伸长,导致细胞死亡[1][2]。 9-β-D-Arabinofuranosylguanine 对培养的 T 淋巴母细胞具有选择性毒性,因为相对于 B- 和无淋巴母细胞,它们能够积累更高水平的细胞毒性代谢物 Ara-GTP[1]。 9-β-D-Arabinofuranosylguanine (0-1000 μM; 72 h) 具有细胞毒性,对 MOLT-4、MOLT-4/Ara-G500 和 MOLT-4/Ara-G900 的 IC50 分别为 4.2、452 和 777 μM[2]。 Cell Viability Assay[2] Cell Line: MOLT-4 and Ara-G-resistant sublines: MOLT-4/Ara-G500 and MOLT-4/Ara-G900 Concentration: 0-1000 μM Incubation Time: 72 h Result: Showed cytotoxicity with IC50s of 4.2, 452 and 777 μM against MOLT-4, MOLT-4/Ara-G500 and MOLT-4/Ara-G900 cells, respectively.
