[体外研究]

当暴露于适当波长的光时经历大的构象变化的光致变色化合物作为分子开关元件特别有吸引力。偶氮苯是发色团中的热门选择。热力学上有利的反式异构体通过在π-π*跃迁的波长下照射而快速转化为顺式异构体,而反向过程通过在黑暗中的热弛豫(缓慢)或通过波长照射(快速)实现。 n-π*过渡的偶氮苯氨基酸(aa)可用作多肽中的光诱导构象转换。通过用合适波长的光照射[1],通过在偶氮苯部分的顺式和反式构型之间切换,诱导肽骨架的可逆构象变化。偶氮苯是用于合成光响应系统的最广泛使用的光学触发剂,从聚α-氨基酸到具有光控机械和光学性质的创新材料。其以适当衍生物的形式使用允许产生约束构象空间的环肽结构,并因此利用其可逆的光异构化来诱导不同构象状态之间明确定义的转换[2]。偶氮苯光电开关可用于驱动肽,蛋白质,核酸,脂类和碳水化合物的功能变化[3]。

偶氮苯制备

由硝基苯按下面反应而得。将硝基苯、甲醇及氢氧化钠溶液加热保持76℃,在搅拌下慢慢加锌粉,保温搅拌16h;再加入锌粉,保温反应48h。趁热过滤,冷却,在粗品中加入20%盐酸在70℃下溶化后,冷却过滤,将结晶物在水浴中加热70-80℃,放冷过滤,最后经用乙醇重结晶、干燥而得。
