壬二酸制备

1. 生产方法简述：由油酸经硝酸氧化或臭氧氧化而得。也可以用工业蓖麻作原料裂解而得。1,5-二溴戊烷与乙腈缩合，或者以戊二酸单甲酯单酰氰为原料经酮式壬二酸二甲酯都可以制取壬二酸。

2. 烟草：OR，44；BU，9；OR，41；BU，26。

3.制法：

于装有搅拌器、回流冷凝器的反应瓶中，加入氢氧化钾100g，95%的乙醇1000mL，蓖麻油500g，搅拌回流3h，将反应液倒入3L水中，以100mL浓硫酸与300mL水配成的稀酸酸化。分出油层，热水洗涤两次，无水硫酸镁干燥。滤去干燥剂，得粗品12-羟基十八烯-9-酸（2）约480g。本品溶液聚合，因而不宜久置。最好及时用于下一步反应。于装有搅拌器、温度计、回流冷凝器的反应瓶中，加入高锰酸钾625g（3.5mol），水7.5L，于35℃剧烈搅拌溶解。一次加入由12-羟基十八烯-9-酸（2）240g（0.8mol）溶于1.6L水和64g氢氧化钾配成的溶液。反应放热，迅速升温至75℃左右。继续搅拌反应0.5h。反应过程中会产生大量泡沫，必要时可加入消泡剂。趁热过量除去MnO2，滤饼用热水洗涤两次。滤液用稀硫酸酸化。减压浓缩至约2L，冰箱中冷却析晶。过滤析出的固体，冷水洗涤，干燥后得壬二酸（1）35～38g.将其用15倍的热水重结晶，活性炭脱色，冷后滤出结晶，干燥，得纯品壬二酸（1）23～27g，mp104～106℃，收率31%～35%。
