[体外研究]

一氧化氮是自由基信号分子。有多种方法可用于测量NO。在所有方法中,用于定位NO的荧光探针是非常广泛使用的方法。二乙酸盐形式的二氨基荧光素(DAF 2DA)是用于NO测量的最广泛的探针。该方法基于4,5-二氨基荧光素二乙酸酯(DAF 2DA)的应用,其活跃地扩散到细胞中,一旦被细胞吸收,细胞质酯酶切割乙酸酯基团以产生4,5-二氨基荧光素; DAF-2。产生的DAF-2可以容易地与N 2 O 3反应,N 2 O 3是NO的氧化产物,以产生高度荧光的DAF-2T(三唑基uorescein)。
