[体外研究]

吖啶橙已被广泛用作细胞化学染色,并且已经显示出差异地染色DNA和RNA,以及原位双重重链核酸。吖啶橙可嵌入双螺旋核酸(530nm处的绿色荧光),或静电结合至单链分子的磷酸基团(640nm处的红色荧光)。这种独特的特性使得吖啶橙可用于细胞周期研究[1]。未固定细胞的吖啶橙染色可用作从人类和小鼠的DNA测量(绿色荧光)和RNA染色(红色荧光)的细胞转录活性获得细胞倍性水平和细胞周期状态信息的简单快速方法。来自白血病和有丝分裂(植物血凝素)或抗原性(混合淋巴细胞培养)患者的细胞系,外周血和骨髓标本刺激人外周血培养[2]。

[细胞实验]

两步，pH3.0：从培养物中取出等分试样（0.2mL，含有约2-5×10 5个细胞）并加入到0.5mL含有：0.1％（v / v）Triton X-100,0.2M蔗糖的溶液中， 10-4M EDTA和2×10-2M柠檬酸盐 - 磷酸盐缓冲液，pH3.0。 Triton X-100包含在指定pH的各种程序中，以增加细胞渗透性，同时保持细胞完整性。螯合剂EDTA用于促进RNA变性。 1分钟后，通过加入1mL含有0.002％（20μg/ mL）AO，0.1M NaCl和10-2M柠檬酸盐 - 磷酸盐缓冲液（pH3.8）的溶液对细胞染色。染料混合物中包含阳离子以确保染色特异性。最终的AO浓度约为4×10-5M [2]。
