胰蛋白酶用途

胰蛋白酶是一种水解赖氨酸或精氨酸羧基侧蛋白质的酶。胰蛋白酶激活PAR2和PAR4。胰蛋白酶通过PDCoV和pAPN的S糖蛋白的相互作用诱导PDCoV感染中的细胞与细胞膜融合。胰蛋白酶也促进细胞增殖和分化。胰蛋白酶可用于伤口愈合和神经源性炎症的研究[1][2][3][4][6]。

[体外研究]

胰蛋白酶(5μg/mL,24或48小时)可促进猪deltacoronavirus(PDCoV)在LLC-PK细胞中的复制[2]。胰蛋白酶(10和50 ng/mL,12小时)通过促进LLC-PK细胞的膜融合,增强了PDCoV在LLC-PK-细胞中的细胞间传播[2]。胰蛋白酶(0.05%,3小时)在无血清和无生长因子培养基中促进C6胶质瘤细胞增殖[3]。胰蛋白酶(20-150 ng/mL,5天)增强PBMC分化[4]。Western Blot分析[2]细胞系：LLC-PK细胞,ST细胞浓度：5μg/mL孵育时间：24或48小时结果：促进了LLC-PK细胞中PDCoV的复制,但不促进ST细胞。免疫荧光[2]细胞株：LLC-PK细胞,ST细胞浓度：10和50 ng/mL孵育时间：12 h结果：在PDCoV感染LLC-PK细胞期间,细胞间融合活性显著增加。

[体内研究]

胰蛋白酶(100-500 50μg/站点 μL生理盐水,皮内注射)诱导小鼠抓挠行为[5]。动物模型：瑞士小鼠[5]剂量：100-500 μg/部位,生理盐水(50 μL)给药：皮内注射结果：引起瘙痒,并被胰蛋白酶抑制剂抑制。
