[体外研究]

AS-252424还抑制PI3Kα,PI3Kβ和PI3Kδ,IC50分别为935±150 nM,20μM和20μM。 AS-252424以浓度依赖性方式抑制MCP-1介导的野生型原代单核细胞趋化性,IC50值为52μM,以及IC50值为53μM的单核细胞系THP-1。在人单核细胞系THP-1中,MCP-1与GPCR趋化因子受体CCR2的结合强烈诱导PKB/Akt的磷酸化,其在低至0.4μM的IC 50值下被AS-252424有效抑制。相反,通过集落刺激因子(CSF-1)诱导PKB/Akt磷酸化,结合生长因子受体c-fms,仅被AS-252424阻断,IC50值高达4.7μM[1]。

[体内研究]

[激酶实验]

基于新霉素包被的闪烁亲近测定（SPA）珠子技术的PI3Kγ脂质激酶测定在384孔板中使用ATP / [γ33P] ATP和PtdIns进行。用PI3Kα，PI3Kβ和PI3Kδ进行IC50值测定的激酶测定[1]。

[细胞实验]

在无血清培养基中饥饿3小时后，用抑制剂（例如，AS-252424,1nM，10nM，100nM，1μM，10μM和100μM）或DMSO预处理Raw-264巨噬细胞30分钟并且用50nM C5a刺激5分钟。使用磷酸-Ser-473 Akt特异性抗体和标准ELISA方案监测PKB / Akt磷酸化[1]。
