[In Vitro]

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs)[1]. 1. The protein to be tested and the protein standard were prepared in 100 mM sodium borate buffer, pH 9.3. 2. Transfer the protein to the wells in amounts from 10 ng to the desired end point within the chosen range. 3. Add the sodium borate buffer to obtain 80 mL total volume. 4. Other buffer (pH 7.2) is PBS, add Triton X-100 to the buffer at the final concentration of 0.1%. 5. A 10-mL volume of 10 mM KCN dissolved in water is added to each well followed by 10 mL of 5 mM CBQCA (prepared immediately before reaction) in sodium borate buffer, pH 9.3, containing 12.5% DMSO. 6. Shake the plates to mix the contents of the wells, protected from light, and incubate for 1-5 h with shaking. 7. Incubate the samples for 90 min. 8. Measure the fluorescence intensity by using the CytoFluorfluorescence plate reader, Ex=488 nm, Em=530 nm.
