Biological Activity

[Description]:

Astramembrangenin (Cycloastragenol) is a triterpenoid saponin compound and a hydrolysis product of the main active ingredient in Astragalus membranaceus (Fisch.) Bunge[1].Astramembrangenin is orally safe and has broad Extensive pharmacological effects, including telomerase activation, telomere elongation, anti-inflammatory and anti-oxidative properties[2].Astramembrangenin has antiaging properties, CAG stimulates telomerase activity in human neonatal keratinocytes and rat neuronal cells, and induces CREB activation followed by tert and bcl2 expression. Cycloastragenol (CAG) may have a novel therapeutic role in depression[2].

[In Vitro]

Astramembrangenin (0-10 µM; 3-6 days) increases cell growth compared to the vehicle control in HEK cultures[1]. Astramembrangenin (0.3 µM; 5-90 mins) induces CREB phosphorylation in primary cortical neurons, total CREB expression is unaltered by CAG in both cell type[1]. Astramembrangenin (3 µM; 6-48 hours) may promotes prosurvival signaling in neurons, increases tert mRNA expression, exhibits increased bcl2 mRNA expression[1]. Cell Viability Assay[1] Cell Line: HEK cells Concentration: 1 µM, 3 µM, 10 µM Incubation Time: 3-6 days Result: Doubled the cell growth at 6 days. Western Blot Analysis[1] Cell Line: Neuronal cells Concentration: 0.3 µM Incubation Time: 5 mins, 15 mins, 30 mins, 90 mins Result: Suggested that CAG induces CREB activation in neuronal cells. RT-PCR[1] Cell Line: Neuronal cells Concentration: 3 µM Incubation Time: 6-48 hours Result: Increased tert, bcl2 mRNA expression.
