Use of NSC 145366NSC 145366 is a novel, allosteric Hsp90 CTD (C-terminal domain) inhibitor, exhibits anti-proliferative activity in tumor cell lines with GI50 of 0.2-1.9 uM; increases Hsp90 oligomerization resulting in allosteric inhibition of NTD ATPase activity (IC50=119 uM), but does not compete with NTD or CTD-ATP binding; causes selective client protein downregulation including AR and BRCA1 but without a heat shock response in LNCaP prostate tumor cells. |
