Target |
STAT3 |
In Vitro | Stattic inhibits STAT3 phosphorylation (Y705) in ALDH+ and D44+/CD24+ subpopulations of Panc-1 and HPAC pancreatic cancer cell lines at a higher concenration (20 μM). Stattic selectively inhibits P-STAT3 as demonstrated by the lack of inhibition of P-ERK1/2 in both ALDH+ and CD44+/CD24+ subpopulations of Panc-1 and HPAC. Stattic inhibita STAT3 downstream targets in ALDH+ and CD44+/CD24+ subpopulations of pancreatic cancer cells[1]. Stattic reduces the LPS-induced expression of ICAM-1 and VCAM-1, and STAT3 phosphorylation[2]. |
Cell Assay | Pancreatic cancer stem-like cells (3,000/well in 96-well plates) are incubated with desired concentrations of compounds in triplicate at 37˚C for 72 h. MTT viability assays are done and the absorbance is read at 595 nm. |

6-Mercaptopurine reduces macrophage activation and gut epithelium proliferation through inhibition of GTPase Rac1.
Inflamm. Bowel Dis. 20(9) , 1487-95, (2014) Inflammatory bowel disease is characterized by chronic intestinal inflammation. Azathioprine and its metabolite 6-mercaptopurine (6-MP) are effective immunosuppressive drugs that are widely used in pa... |

Cigarette smoke-induced MMP2 and MMP9 secretion from aortic vascular smooth cells is mediated via the Jak/Stat pathway.
Hum. Pathol. 46(2) , 284-94, (2015) It is hypothesized that cigarette smoke may increase MMP2 and MMP9 secretion through Jak/Stat pathway in the aorta, thereby facilitating abdominal aortic aneurysm (AAA) formation/progression in smoker... |

Reduction of RKIP expression promotes nasopharyngeal carcinoma invasion and metastasis by activating Stat3 signaling.
Oncotarget 6 , 16422-36, (2015) The role and underlying mechanism of Raf kinase inhibitory protein (RKIP) in nasopharyngeal carcinoma (NPC) metastasis remain unclear. Here, we showed that RKIP was downregulated in the NPC with high ... |
