Kinase Assay | Fluorescence measurements are carried out using a Cary Eclipse fluorescence spectrophotometer equipped with 1.0 cm quartz cells. Prior to fluorescence measurements, an appropriate volume of Safflower red solution was added to HSA (2.0 μM), ranging from 0 μM to 0.3 μM, and mixed for 30 min at 25°C. The protein intrinsic fluorescence of HSA and HSA-Safflower red systems were measured at an excitation of 280 nm. Emission spectra were recorded from 300 nm to 500 nm, with widths of excitation and emission slits set at 5 and 10 nm, respectively. All fluorescence intensities were corrected to decrease the inner filter effect[1]. |
