In Vitro Immunoprecipitated FLAG-dDYRK2 or FLAG-MNB is assayed using 50 μM Woodtide or 50 μM activation-loop peptide with two additional lysines attached to the N terminus to allow it to bind to P81 paper (KKSSCYVDRKIYTYIQSRFY for dDYRK2 or KKSSCQLGQRIYHYIQSRFY for MNB). Both kinases efficiently phosphorylated the synthetic exogenous substrate peptide Woodtide, but neither dDYRK2 nor MNB phosphorylated their activation-loop peptide[1].
