Processing and activation of procathepsin S[2]: 1. Autocatalytic activation of procathepsin S was studied by incubation (final concentration 1-5 μM) at 37 ℃ in 0.5 mL of the appropriate buffer containing 2.5 mM dithiothreitol. 2. Aliquots of 10 μl were taken from the reaction mixture at the times indicated and mixed with 2.5 ml of substrate solution (10 μM Bz-FVR-AMC in 0.1 M phosphate buffer, pH 6.5, containing 1 mM EDTA (HY-Y0682) and 0.1% (w/v) polyethylene glycol 6000). 3. Fluorescence of the released AMC was then monitored continuously for 1 min in a pectrofluorimeter at excitation and emission wavelengths of 370 and 460 nm.
