[体外研究]

BCECF-AM用于测量肺动脉平滑肌细胞(PASMC)中ET-1浓度(0.1-10 nM)增加诱导的基础pHi和NHE活性的变化[1]。

[细胞实验]

将PASMC置于用HBSS灌注的层流式细胞室中，pH调节至7.4。在pH敏感性荧光染料BCECF-AM的膜渗透物（乙酰氧基甲基酯）形式与37℃，20％O 2 -5％CO 2气氛下孵育60分钟的细胞中测量pH 1。然后将细胞用HBSS在37℃洗涤15分钟以除去细胞外染料并使细胞溶质染料完全脱酯化。 BCECF荧光的比率测量在由具有落射荧光附件的Nikon TSE 100 Ellipse倒置显微镜组成的工作站上进行。来自氙弧灯的光束通过490和440nm的干涉滤光器过滤，并通过20倍荧光物镜聚焦到检查中的PASMCS上。从530nm处的电池发出的光通过物镜返回并由成像相机检测。电子快门用于最小化染料的光漂白。执行协议并使用InCyte软件在线收集数据。在每次实验后通过原位校准估计pHi。用含有（以mM计）：105KCl，1MgCl 2,1.5 CaCl 2,10葡萄糖，20HEPES-Tris和0.01尼日利嗪的溶液灌注细胞，以使pHi平衡至外部pH。当用KOH调节pHi从6.5到7.5时，测量荧光产生两点校准。细胞内H +离子浓度（[H +] i）由pHi使用下式确定：pHi = -log（[H +] i）。
