4-甲基伞形烯醇油酸酯是酸性和碱性脂肪酶的荧光底物。4-甲基伞形烯丙基油酸酯被脂肪酶裂解,释放出4-甲基伞状烯丙基(Ex/Em=320/450 nm)[1][2]。

[体外研究]

以4-甲基伞形科(4-MU)油酸酯为底物,测定其对胰脂肪酶的抑制活性。反应混合物由50µL 0.1 mM 4-MU油酸盐、20µL McIlvane缓冲液(0.1 M柠檬酸-Na2 HPO4,pH 7.4)和5µL样品溶液组成。将猪胰脂肪酶(25µL)添加到反应混合物中,并将最终体积调整为0.1 ml。在混合物在37℃孵育10 min后,使用荧光多重检测仪在320 nm激发波长和450 nm发射波长下测量脂肪酶释放的4-MU量[1]。
