[体外研究]

Cynarin抑制味觉受体,使水变甜。它已被证明具有一些药理学性质,包括降胆固醇,保肝,抗病毒,抗菌和抗组胺作用。 Cynarin具有显着的抗氧化,抗胆碱能,还原能力,自由基清除和金属结合活性。在30mg/mL浓度下,Cynarin表现出87.72％的亚油酸脂质过氧化抑制。 Cynarin表现出有效的DMPD +,ABTS +/sup>,O2-,DPPH1和H2O2清除作用,降低能力和Fe2 +螯合作用。 cynarin对乙酰胆碱酯酶抑制的IC50和Ki分别为243.67nM和39.34±13.88nM [1]。 Cynarin是一种潜在的免疫抑制剂,可阻断T细胞受体的CD28与抗原呈递细胞的CD80之间的相互作用。在1比1的T细胞和B细胞比例的条件下,Cynarin阻断了约87％的CD28依赖性“信号2”T细胞活化途径。 Cynarin与CD28的“G-pocket”结合,从而中断CD28与CD80之间的相互作用位点[2]。

[细胞实验]

通过MTT比色测定法测量cynarin处理T细胞的细胞毒性。将100μLJurkat细胞与cynarin（0-1000μg/ mL）在37℃下孵育24小时。然后离心细胞溶液并除去上清液。加入200μLMTT，将细胞溶液再次在37℃温育4小时。将200μLDMSO裂解缓冲液加入细胞培养基中，用读板仪在560nm处测量溶解的MTT晶体的浓度[2]。
