[体外研究]

Hygromycin B是一种强烈抑制70S和80S核糖体的氨基环醇抗生素,由吸水链霉菌(Streptomyces hygroscopicus)合成[1]。 0.38mM浓度的潮霉素B完全阻止富含培养基中的酵母细胞生长,可能是通过细胞质核糖体阻止蛋白质合成。来自兔网织红细胞,小麦胚芽和酵母的无细胞提取物中的多肽合成被低浓度的潮霉素B强烈阻断。抗生素通过预防延伸因子EF-2依赖性易位来抑制酵母多核糖体的肽链延伸。潮霉素B对易位的抑制可能是由于与核糖体受体位点结合的肽基-tRNA的稳定作用[2]。

[体内研究]

潮霉素B通过阻断核糖体易位来抑制蛋白质合成,而不会在体内引起显着的误读[3]。在体内转基因小鼠中细菌hygR基因的组成型表达赋予对潮霉素B的抗性[4]。

[动物实验]

将潮霉素B溶于无菌水中。携带hygR的小鼠C57BL / 6J-TgN（pPWL512hyg）1Ems用单剂量的潮霉素B ip处理，剂量开始为2.7mg / kg，并且每次连续剂量增加50％。用相同体积的无菌盐水处理对照野生型C57BL / 6J小鼠。注射的总体积为0.5mL。每天监测动物的健康状况和体重，连续10天[4]。
