[体外研究]

吡罗昔康是一种非甾体类抗炎药,作为COX抑制剂,人单核细胞COX-1和COX-2的IC50分别为47,25μM[1]。吡罗昔康(167,333,500μM)减少T24和5637细胞的细胞群。吡罗昔康(500μM)也降低了T24和5637细胞的细胞活力,并且当与0.05μM卡铂组合时显着有效。该组合还抑制了展位细胞中Ki-67的表达[3]。

[体内研究]

Piroxicam(0.3 mg/kg qd 24-h po)可降低18只狗中12只的肿瘤体积,这种效果是通过诱导细胞凋亡和降低尿液碱性成纤维细胞生长因子浓度来实现的[2]。

[细胞实验]

用分级浓度的卡铂(0.05,0.5和1μM)和吡罗昔康(167,333和500μM)处理膀胱癌细胞系72小时以评估剂量 - 反应谱。对于组合方法,使用0.05μM卡铂和333μM吡罗昔康。在每次实验之前,两种药物都是新鲜制备的。未处理的对照组(未暴露于卡铂和吡罗昔康的细胞)用于所有测定[3]。

[动物实验]

狗[2]狗在治疗前和吡罗昔康(0.3 mg / kg qd 24-h po)治疗4周后接受肿瘤分期,包括胸部和腹部X线摄影,膀胱造影,超声检查和膀胱镜检查(收集组织样本)。在吡罗昔康治疗的4周期间,狗没有接受任何其他癌症治疗。将组织样品立即在液氮中冷冻用于PGE2分析或固定在10%中性缓冲福尔马林中用于免疫组织化学检查。在吡罗昔康治疗之前和之后也收集尿液,等分,然后储存在-80℃直至分析[2]。
