三环唑制备
1.以邻甲苯胺等为原料,经加成、关环、取代、扩环四步反应制得三环唑。
加成反应 将邻甲苯胺在溶剂存在下加入硫酸,再加入硫氰化钠,升温反应。经回收溶剂、冷却、过滤、干燥等后处理,得邻甲基苯基硫脲,m.p.154 ~160℃,收率93%~95%。
关环反应 在溶剂存在下,将邻甲基苯基硫脲与氯反应,注意控制反应温度。保温反应结束后,回收溶剂、加碱中和、冷却、过滤、甩干,得2-氨基-4-甲基苯并噻唑。m.p.134~136℃,收率90%~91%。
取代反应 将适量的溶剂、盐酸和水混合后,加入2-氨基-4-甲基苯并噻唑,与肼的盐酸盐作用,升温反应至终点,冷却、过滤、干燥后得2-肼基-4-甲基苯并噻唑。m.p. 167~170℃,收率85%~87%。
扩环反应 将2-肼基-4-甲基苯并噻唑在搅拌下加入到一定量溶剂和甲酸的反应釜中,加热升温至反应结束、减压回收溶剂和过量的甲酸,同时加水稀释冷却,过滤、水洗至中性,甩干、干燥得三环唑原药。m.p. 184~186℃,收率90%~93%。
2.将邻甲苯胺在溶剂存在下加入硫酸,再加入硫氰化钠,升温反应,经处理得邻甲基苯基硫脲。在溶剂存在下将邻甲基苯基硫脲与氯气反应,经加碱中和、冷却、过滤、甩干,得2-氨基-4-甲基苯并噻唑。将适量的溶剂、盐酸和水混合后,加入2-氨基-4-甲基苯并噻唑,与肼的盐酸盐作用,升温反应至终点,冷却、过滤、干燥后得2-肼基-4-甲基苯并噻唑。然后将2-肼基-4-甲基苯并噻唑在搅拌下加入到一定溶剂和甲酸中,加热升温至反应结束,减压回收溶剂和过量的甲酸,同时加水稀释冷却、过滤、水洗至中性,甩干、干燥得三环唑原药。
