斑蝥素制备
1.斑蝥素是从鞘翅类地胆科斑蝥属昆虫斑蝥中提取的一种单萜烯类物质,成虫中含量为1%左右。
首先将斑蝥粉碎成粉状。按斑蝥虫粗粉∶浓盐酸∶丙酮=1∶0.025∶5.5的比例投料。取干燥粗粉加入浓盐酸,然后加入3倍量的丙酮,浸泡提取48h,然后用双层纱布过滤,收集滤液,残渣再用25倍量丙酮浸泡48h,过滤,收集提取液,合并提取液,残渣弃去。回收提取液中的丙酮,得到深色浓缩油状物,油状物中有大量结晶析出,过滤,收集结晶。结晶用石油醚洗涤两次,除去色素和动物脂肪,再用95%乙醇洗涤数次,干燥,得斑蝥素粗品。粗品用丙酮再结晶一次,即得斑蝥素。斑蝥素有剧毒,制备时要注意防护,绿茶和黄连等可解毒。
2. 本品是将昆虫斑蝥 [ Epicauta gorhami marseul(Meloidae)]干燥后用乙醇浸出制得的酊剂。欲制本品1000ml,先将斑蝥干燥,取粉碎的粗末100g,加乙醇约600ml,不时搅拌,让可溶性成分充分溶出,用布过滤。然后将残留物用少量乙醇洗涤并压干,合并浸出液及洗液,放置2天后过滤,滤液中再加乙醇至1000ml即成。
