[体外研究]

OtBu MTX PVG比MTX-PVG对HT1080细胞的HT1080更小。这表明MTXα-羧基的保护导致体外活性降低[1]。

[体内研究]

Lys-mtxcleable-(OtBu)显著降低HT1080荷瘤小鼠的肿瘤生长[1]。甲氨蝶呤α-羧基的保护可用于改善类似MTX结合树状大分子的循环半衰期和减少肝脏蓄积,同时仍保持体内抗肿瘤活性[1]。动物模型：雌性,Balb/c nu/nu,6周(携带HT1080细胞)[1]剂量：30mg/kg给药：每周1次,持续2周。结果：第12天肿瘤体积显著减小(63%)。
