细胞实验 | 将细胞(每孔5×103个细胞)在96孔板中培养过夜以进行附着,洗涤,然后在溶液中固定。将等分试样(100μL)的反应缓冲液(即,没有X-Gal的染色溶液)加入每个孔中。然后,每孔加入10μL2mM荧光素二(β-D-吡喃半乳糖苷),将板在黑暗中于37℃温育24小时,不加入CO 2。在37℃温育24小时后,将100μL上清液转移到96孔板中,一式三份进行荧光测量。使用荧光计测量荧光素荧光,激发波长为485nm,发射波长为535nm [1]。 |
