In Vitro | Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs). Labeling of Cells: 1. Fresh N. oceanica cultures are diluted to ~4×106 cells/ml with ASW and kept at 22℃ prior to any treatment. Incubate the cells according to your normal protocol. 2. BODIPY 505/515 is dissolved in DMSO at 4 mg/ml and diluted with DMSO to different working stock concentrations. 3. Cell suspensions are supplemented with the appropriate BODIPY 505/515 working stock to a specific DMSO concentration between 2 and 10% (v/v) with final BODIPY concentrations between 0.8 and 4 μg/ml. 4. Pure DMSO was used for control treatments. 1 ml of fresh culture was diluted to ~4×106 cells/ml with ASW and stained with 6% DMSO and 1.2 μg/ml BODIPY for 15 min (non-stressed cultures) or with 10% DMSO and 1.6 μg/ml BODIPY for 36 min (stressed cultures). 5. Upon addition of the dye, samples were vortexed for 5 s and then incubated in the dark for 15 min before flow cytometric analysis, if not indicated otherwise. |
