Description | GTC-365 (NSC 177365) is a small drug-like pharmacological chaperone that induces cancer cell death by restoring tertiary DNA structures in mutant human telomerase reverse transcriptase (hTERT) promoters; acts at an early step in the G-quadruplex folding pathway to redirect mutant promoter G-quadruplex misfolding, partially reinstates the correct folding pathway, and reduces hTERT activity through transcriptional repression; binds to WT and G124/125A telomeric G-quadruplexes with a similar binding affinity (Kd=400 nM). |
