Kinase Assay | 5 μL of a solution of IRAK inhibitor 3 at a concentration of 50 μM or less in 1% (v/v) DMSO is added to the wells of a 96-well area Black Polystyrene plates. The final concentrations in the reaction well are 10 μM ATP, 0.5 μM IRAK4 CH373, 1.6 μM IRAKI peptide, 1 mM MgCl2, 2 mM DTT, 5 mM MnCl2, 0.01 % BSA, and 0.01 % Tween-20. The volume of the reaction is 45 μL. The reaction mixture is incubated at room temperature for 30 minutes and stopped with the addition of 5 μL of 100 mM EDTA[1]. |
