In Vitro Guidelines[2] (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs). Labeling of Human Sperm Cells: 1. Incubate the cells according to your normal protocol. 2. Load cells with 5 μM Fura-FF in 1 mL Hepes-buffered human sperm (H-HSM) medium at 36 ℃ for 40 min. 3. Wash the sample by centrifugation and scatter cells in disposable centrifuge tubes at 1 ◊ 107 cells in 3 mL of H-HSM and maintained at 36 ℃. 4. Immediately measure fluorescence recordings.
