[体外研究]

通过Western印迹测定,香草酸抑制脂多糖(LPS)诱导的细胞质提取物中IκB(细胞质NF-κB抑制剂)的磷酸化和体外巨噬细胞核提取物中NF-κBp65亚基的表达[1]。香草酸处理可逆转Aβ1-42诱导的IKK-β/ NF-κB表达升高,并降低IKKβ/ NF-κB的活性。在所有三种浓度(50,100和200μM)下,香草酸对HT22细胞无毒;与Aβ1-42的香草酸共处理显着增加(分别为1.5倍,1.9倍和2倍)细胞活力。与对照细胞相比,Aβ1-42(5μM)处理导致ROS水平显着增加(1.6倍)。相比之下,所有三种不同浓度(50,100和200μM)的香草酸处理均降低(分别为1.1倍,1.3倍和1.4倍)ROS水平,表明香草酸是一种有效的抗氧化剂[2]。

[体内研究]

香草酸(30mg/kg,ip,1h)抑制角叉菜胶诱导的NF-κB活化。小鼠在ip注射角叉菜胶(300μg/爪)前1小时接受香草酸(30mg/kg,ip),并在刺激注射后3小时收集皮肤足底组织的样品。卡拉胶诱导的NF-κB活化通过总NF-κB/ p-NF-κB比例降低来观察,而用香草酸和对照药物吲哚美辛(5 mg/kg,ip,稀释)进行治疗。刺激前40分钟的Tris/HCl缓冲液抑制角叉菜胶诱导的NF-κB活化[1]。

[细胞实验]

用MTT测定评估HT22细胞的活力。将细胞在96孔板中以每孔1×10 4个细胞的密度在100μLDulbecco改良的Eagle培养基（DMEM）中培养。 24小时后，用含有Aβ1-42（5μM）的新鲜培养基替换培养基，其中三种不同浓度（50,100和200μM）的香草酸单独或与Aβ1-42（5μM）组合，并且将细胞再孵育24小时。对照细胞仅接收DMEM。此后，将细胞与MTT溶液在37℃下再孵育2-4小时。随后，用DMSO替换每个孔中的培养基。最后，使用ApoTox仪器测量570nm处每个孔中溶液的吸光度。所有实验均一式三份进行[2]。

[动物实验]

小鼠[1]在实验过程中，雄性瑞士小鼠（25-30g）在给予香草酸或载体（5％吐温80稀释于盐水中）前1小时或24小时接受腹膜内（3,10或30mg / kg）治疗。 h炎症刺激注射后[1]。
