[体内研究]

向PND9小鼠单次全身注射L-谷氨酸单钠盐(MSG)(3μg/ kg)引起广泛的视网膜变性,并且相对于用盐水处理的对照小鼠,Brn-3a + RGC的数量减少> 70%;在用L-谷氨酸单钠盐和JNJ16259685处理的小鼠中,RGC的数量仅减少<20%,并且得到的值与包括对照小鼠在内的所有其他组小鼠中获得的值没有显着差异。向野生型BALB/c同窝仔畜施用L-谷氨酸单钠盐可使RGC数量降低至低于C57BL/6J小鼠的程度,但降低仍然非常显着[1]。

[动物实验]

对于毒性实验,如下处理PND9小鼠(每组n = 3-9只小鼠)。两组接受皮下(sc)注射玉米油,30分钟后,通过皮下注射生理盐水或L-谷氨酸单钠盐(3g / kg);用3,4-二氢-2H-吡喃并[2,3-b]喹啉-7-基 - (顺式-4-甲氧基环己基) - 甲酮处理另外两组,溶于玉米油(2.5mg / kg)中。 30分钟后,通过皮下注射生理盐水或L-谷氨酸单钠盐。在另一组实验中,用盐水或L-谷氨酸单钠盐(3g / kg)皮下注射四组crv4小鼠或其野生型同窝小鼠(每组n = 5-7只小鼠)[1]。

谷氨酸钠制备

1.由谷氨酸中和、精制而得。将谷氨酸溶于水中,用含盐分少的碳酸钠或固体氢氧化钠中和至pH=6.7-7.0,温度不超过60℃。中和液经脱色去铁后进行真空浓缩、结晶、分离干燥而得成品。

目前国内外均采用以大米、淀粉或糖蜜为原料经糖化、发酵、提取和精制等工序制得:
(C6H10O5)N[H2O]→C6H12O6[NH3,O2] →C5H9NO4[Na2CO3] →C5H8NnaO4
淀粉的糖化淀粉的糖化多采用双酶法。大米经浸泡、砂磨或淀粉直接调配成相对密度1.1(14°Bé)的淀粉浆;在Ph值4.5和90℃的条件下加入α-淀粉酶液化15~20min,然后升温至l00℃杀酶5min;放料后经板框压滤,滤渣可作饲料,滤液调至Ph值5.0,加入β-淀粉酶,在60℃下糖化12h;再升温至100℃杀酶后冷却澄清,上层清液直接放入糖液贮槽,沉淀液加助滤剂硅藻土后压滤,淀粉糖转化率可达92%。
谷氨酸发酵以15%左右的葡萄糖为碳源,并加适量的无机盐和生物素配成发酵培养基,经连消并冷却至40℃后送入已灭菌的发酵空罐;以流加的液氨为氮源,接种经二级扩大培养的谷氨酸产生菌,通风发酵30h;糖酸转化率可达50%,产酸水平约7.0%~7.5%。发酵过程Ph值控制在7.0~7.2,温度前期控制在32~34℃,后期34~37℃。
2.谷氨酸的提取谷氨酸的提取现一般采用冷冻等电一离子交换法。发酵液在等电罐中一边用冷冻盐水缓慢搅拌冷却降温至5"C,一边用硫酸调Ph值至3.22(等电点);沉淀8h后,沉淀经离心分离得粗谷氨酸;母液和上层清液调配后上离子交换树脂交换,用氨水洗脱;前流分汇入上层清液重新上柱,后流分与氨水一起作洗脱液,高流分与发酵液一起回等电罐;提取收率可达88%~90%。
谷氨酸精制 在装有60~6 5℃底水的中和罐中加入谷氨酸,搅拌,并缓慢加入纯碱溶液,中和至Ph值6.2~6.4,中和液浓度控制在相对密度1.1 7~1.18(21~2 2°Bé);待中和液降温至50℃以下,加入适量的硫化钠溶液以除铁;然后用粗谷氨酸回调Ph值至6.2~6.4,并升温至60℃,再加入粉末活性炭,搅拌半小时后送入压滤机压滤;再将滤液用颗粒活性炭柱二次脱色得清液;清液送入真空煮晶锅内在60~70℃下蒸发浓缩至相对密度1.28(31.5084),加入0.3 6~0.542mm的晶种后继续蒸发结晶,期间需用热水杀晶和补加一定量的清液;放料后,经育晶槽,再离心分离得结晶味精,母液或经脱色后再蒸发结晶,精制收率可达理论量的92%。

3.由发酵所得L-谷氨酸与氢氧化钠或碳酸钠中和后脱色精制而得。
