[体外研究]

Xenin在胃,十二指肠和空肠粘膜中大量表达,并且在胰腺中发现较低水平。据报道,Xenin在餐后循环释放,刺激胰腺内分泌和外分泌,抑制胃泌素分泌,影响胃肠动力。 Xenin与神经降压素高度同源。据报道,Xenin和神经降压素具有相似的生物学作用[1]。

[体内研究]

脑室内和腹膜内施用xenin均以剂量依赖性方式抑制野生型小鼠中的空腹诱导的食欲过盛。腹膜内注射xenin还可减少自由采食的野生型小鼠的夜间摄入。腹膜内注射xenin可增加下丘脑核中的Fos免疫反应性,包括室旁核和弓状核。 Xenin减少了agouti和ob/ob小鼠的食物摄入[2]。与盐水处理的对照小鼠相比,在xenin处理的小鼠中胃排空率降低约93％。 ip xenin注射显着增加脑干孤束核内的Fos免疫反应细胞,但不能增加迷走神经区后区和背侧运动核[3]。

[动物实验]

小鼠：小鼠（野生型和ob / ob小鼠）禁食过夜（1800-1000小时）并脑室内注射xenin（0.1,1或5μg）或腹膜内注射（0.5,5,15或50μg/ g）体重wt）在1000小时。为了比较xenin和神经降压素之间的进食抑制作用，在禁食过夜后腹膜内注射等摩尔量（16.5nmol）的xenin（50μg/ g体重）或神经降压素（28μg/ g体）。对照小鼠接受脑室内注射aCSF或腹膜内注射盐水。在注射后立即向小鼠提供预先称重的食物。在注射后24小时内，每个图中所示的时间点测量累积食物摄入量[2]。
