.将新鲜或冷冻的胸腺除脂肪并绞碎后，加3倍量生理盐水，于组织捣碎机中制成匀浆，高速离心后得提取液（组分1）。提取液在80℃加热15min，以沉淀对热不稳定的杂蛋白。离心去掉沉淀，得上清液（组分2）。上清液冷却至4℃，然后加入5倍体积的-10℃丙酮，过滤收集沉淀，干燥后得丙酮粉（组分3）。将丙酮粉溶于pH=70磷酸盐缓冲溶液中，加硫酸铵至饱和度为025，离心去除沉淀，上清液（组分4）调pH=40，再加硫酸铵至饱和度为050，得盐析物。将盐析物溶于pH=8的磷酸盐缓冲溶液，过滤，然后取相对分子质量在15000以下的超滤液。超滤液再经Sephadex G25柱脱盐后，冷冻干燥，可得胸腺素。
