Indatraline hydrochloride用途

盐酸吲达曲林(Lu 19-005)是一种非选择性单胺转运体抑制剂,可阻断神经递质(多巴胺、5-羟色胺和去甲肾上腺素)的再摄取,其功效与可卡因相似。盐酸吲达曲林可用于抗抑郁药物的研究。盐酸吲达曲林诱导自噬,同时抑制细胞增殖。盐酸吲达曲林还可用于指导自噬相关疾病(如动脉粥样硬化或再狭窄)新药物的开发[1]。

[体外研究]

吲达曲林(0~5μM；24小时；EGFP-LC3稳定细胞)盐酸盐使LC3的转化呈浓度依赖性[1]。盐酸吲达曲林(1~10μM；24小时；HeLa细胞)使细胞质中EGFP-LC3荧光液泡浓度依赖性增加[1]。盐酸吲达曲林(1~20μM；平滑肌细胞)抑制平滑肌细胞增殖,IC50为15μM。盐酸吲达曲林诱导细胞自噬。盐酸吲达曲林影响AMPK/mTOR/S6K信号轴[1]。westernblot分析[1]细胞系：HeLa细胞浓度：0~5μM孵育时间：24小时结果：使LC3发生浓度依赖性转化。免疫荧光[1]细胞系：EGFP-LC3稳定细胞浓度：1~10μM孵育时间：24小时结果：EGFP-LC3荧光液泡在细胞质中浓度依赖性增加。

[体内研究]

盐酸吲达曲林(2μM)抑制平滑肌细胞的新内膜积聚[1]。
