体外研究 CGP37157(化合物XVI)是一种有效的选择性Na +/Ca2 +交换抑制剂,可抑制豚鼠心脏线粒体中Na +诱导的Ca2 +释放,IC50为0.8μM[1]。 CGP37157(10μM)显示对皮质神经元中线粒体Na +/Ca2 +交换剂的抑制作用,通过抑制电压门控钙通道调节细胞内Ca2 +水平,并减少NMDA诱导的细胞溶质和线粒体Ca2 +超载。 CGP37157(10μM)也可降低NMDA诱导的兴奋性毒性,这种作用是通过减弱神经元中线粒体损伤和钙蛋白酶活性来实现的[2]。 CGP37157(10μM)与沙利霉素组合显着减弱细胞活力并增加FaDu和HLaC79细胞的凋亡。此外,CGP37157对沙利霉素肿瘤毒性没有抑制作用[3]。
细胞实验 进行细胞毒性测定。根据实验,神经元暴露于含有2.6mM CaCl 2,10mM葡萄糖和10μM甘氨酸的HBSS(不含Ca 2+和Mg 2+)中的NMDA,在37℃下10或30分钟。在兴奋毒性损伤之前和期间存在CGP37157,并且使用Citotox 96比色测定在24小时后评估细胞活力。所有实验均以一式四份进行,所提供的值是至少三次独立实验的归一化平均值±SEM [1]。
