In Vivo | Oral administration of 1-Acetyl-3-o-toluyl-5-fluorouracil demonstrates a remarkable effect on MH134 solid tumors, the effect being more marked than that of subcutaneous administration of 1-Acetyl-3-o-toluyl-5-fluorouracil. Anti-tumor activity of oral administration of 1-Acetyl-3-o-toluyl-5-fluorouracil at a dose of 0.2 mmol/kg/d is comparable to that of subcutaneous administration of 5-fluorouracil at the same dose. The level of decrease in thymus weight and the magnitude of increase of spleen weight following oral administration of 1-Acetyl-3-o-toluyl-5-fluorouracil at any dose are smaller than those by subcutaneous administration of 5-fluorouracil (0.2 mmol/kg/d)[1]. |
Animal Admin | Mice: Of the ten groups of tumor bearing mice, subcutaneous administration of 5-fluorouracil or 1-Acetyl-3-o-toluyl-5-fluorouracil is made at a dose of 0.2, 0.4 and 0.6 mmol/kg/d on six groups in the buttock region. In three groups, oil solution of 1-Acetyl-3-o-toluyl-5-fluorouracil is administered orally with a stomach tube at a dose of 0.2, 0.4 and 0.6 mmol/kg/d. The remaining group is used as tumor bearing control. During the period of the experiment, body weight is measured every 3 day and the long and short diameters of the tumor are measured daily[1]. |
