[体外研究]

剂量依赖性的Galantide(IC50 = 1.0nM)拮抗甘丙肽介导的对小鼠胰岛中葡萄糖诱导的胰岛素分泌的抑制。还发现拮抗剂从产生Rin m 5F细胞的胰岛素的膜中置换125 I-单碘- [TyrZ 6]甘丙肽,IC 50值小于0.1 nM [1]。

[体内研究]

侧脑室注射甘丙肽(5微克/大鼠)抑制有经验的雄性大鼠的性行为-不产生任何其他运动或行为缺陷-注射甘丙肽拮抗剂,甘草,通过相同的途径(1或2微克/大鼠)刺激性行为(改善唤醒,动机和表现指数)和拮抗甘丙肽的作用[2]。 Galantide改善轻度急性胰腺炎(AP)。 Galantide显着降低AP诱导的高酶血症41-49％[3]。已发现Galantide在“社会认知”测试中改善社会记忆,当icv以6-6000 nM的剂量给药时[4]。

[动物实验]

大鼠：将Galantide溶解在0.9％NaCl中。在第一次遭遇结束时，在移除少年后立即给予常驻大鼠icv注射。对照大鼠以相同的途径，以相同的输注速率（1μL/ 20s）同时接受等体积（4μL）的盐水。每个剂量水平至少使用10只成年大鼠[4]。小鼠：Galantide储备溶液在盐水中的0.01％牛血清白蛋白中制备。将小鼠分成7组：给予注射载体的小鼠（仅盐水），仅给予雨蛙素的小鼠（仅AP），仅给予甘草（仅GT）或feG（仅用feG）的小鼠，给予雨蛙素和甘草的小鼠（AP） + GT）或caerulein和feG（AP + feG），最后是施用了caerulein，galantide和feG（AP + GT + feG）的小鼠。在AP诱导前一天，收集血样以测量基础血浆淀粉酶活性[3]。
