体内研究 | 急性给予达索罗林剂量依赖性地分别通过激活α2,D2和5-HT1A自身受体降低LC NE,VTA DA和DR 5-HT神经元的自发放电率。 Dasotraline主要抑制LC NE神经元的放电率,同时仅产生VTA DA和DR 5-HT神经元放电的部分降低。 SEP-225289在抑制5-HT和NE转运蛋白方面具有同等作用,因为它可以延长50%恢复背侧海马CA3锥体神经元的放电活性所需的时间,这是由微电解质应用5-HT和NE [1]。随着SEP-225289剂量的增加,平均多巴胺和血清素转运蛋白占有率增加。对于8mg,平均多巴胺和5-羟色胺转运蛋白占据率分别为33%±11%和2%±13%; 12毫克分别为44%±4%和9%±10%; 16 mg [49]分别为49%±7%和14%±15%。 |
