[体外研究]

氟西汀阻断由海马细胞的不可避免的休克(IS)引起的细胞增殖的下调[1]。氟西汀增加成年大鼠海马齿状回中新生细胞的数量。氟西汀也增加了前肢皮质中增殖细胞的数量[2]。氟西汀加速未成熟神经元的成熟。氟西汀增强齿状回神经发生依赖性长时程增强(LTP)[3]。氟西汀,但不是西酞普兰,氟伏沙明,帕罗西汀和舍曲林,可增加前额皮质中的去甲肾上腺素和多巴胺细胞外水平。急性全身给药后,氟西汀可使去甲肾上腺素和多巴胺的细胞外浓度稳定持续增加[4]。

[体内研究]

氟西汀治疗还可以逆转在成年雄性Sprague-Dawley大鼠中暴露于不可避免的休克的动物中观察到的逃避潜伏期的缺陷[1]。氟西汀(5 mg/kg)单独增加齿状回中的细胞增殖。与对照组相比,共同给药(氟西汀5 mg/kg +奥氮平)也显着增加BrdU阳性细胞的数量[2]。氟西汀联合奥氮平可使细胞外多巴胺水平([DA](ex))和去甲肾上腺素([NE](ex))稳定持续增加,分别达到基线的361%和272%,显着高于基线水平。单独使用药物[5]。

[动物实验]

体重250-300g的雄性Sprague-Dawley大鼠在12小时光照/ 12小时黑暗周期(上午7:00点亮,晚上7点点亮)和恒温(25℃)下饲养。和湿度,允许自由获取食物和水。对于慢性药物治疗,给大鼠施用氟西汀(5mg / kg /天)或盐水通过腹膜内(IP)注射每天一次,奥氮平或载体在饮用水中施用21天(溶媒处理对照,氟西汀和奥氮平)加上氟西汀和奥氮平的组合。对于联合治疗,选择奥氮平是因为已知氟西汀会干扰奥氮平的代谢并使血液水平升高4-6倍。将奥氮平溶解在盐酸(HCl)中,然后用1N氢氧化钠调节回pH6,使浓度为3mg / mL的储备溶液。向对照动物中加入相同量的载体溶液。每周测量三次液体摄入量,并用新鲜药物溶液补充饮用瓶。治疗组之间的液体摄入量没有差异。对于亚慢性治疗,药物以完全相同的方式施用,但总共7天。
