An antimetabolite is a compound similar in structure to a normal metabolite or coenzyme, and its main function is to bind to an enzyme necessary for the synthesis of a normal metabolite, thereby interfering with the synthesis of nucleic acid and inhibiting the growth and proliferation of tumor cells. Some antimetabolites have feedback inhibition that blocks the synthesis of metabolite precursors. The main anti-metabolites are anti-folate, anti-purine and anti-pyrimidine, which act on the S phase of the cells and are all cell cycle-specific drugs. Because there is no obvious qualitative difference between the metabolic process of tumor tissue and normal tissue, the selectivity of antimetabolites is poor, and it is quite toxic to normal tissues with rapid proliferation such as bone marrow, gastrointestinal mucosa, reproductive system and hair. . Although most of the antimetabolites inhibit the synthesis of cellular DNA, there is generally no cross-resistance between the antimetabolites because of their different action points.
